Antibody preservation precautions and storage conditions

Whether the antibody is stored properly or not directly determines the activity and use of the antibody! Most antibodies can last for months or even years if the antibodies are properly stored. The abcam antibody preservation method is mainly used, and it can also be applied to most other antibody products!

1. After receiving the antibody, be sure to centrifuge at 12000 rpm for 1-5 minutes before opening the tube cover for dispensing and storage!

The high-quality antibody uses a very special storage tube, and only the antibody attached to the wall or cap can be centrifuged by centrifugation at 12000 rpm for 1-5 min . Even centrifugation at 10,000 rpm can result in incomplete centrifugation, resulting in an insufficient amount of antibody!

2. For most antibodies, a suitable storage method is to store at -20 degrees or -80 degrees after dispensing . For most antibodies, it is sufficient to store at -20 degrees. There is no evidence that there is more to be saved at -80 degrees!

(1) Dispensing can minimize the damage of antibody activity caused by repeated freezing and thawing, and also reduce the possibility of contamination caused by multiple times of taking antibodies from the same tube.

(2) The amount of dispensing is preferably used up in one experiment, and at least not less than 10ul per serving. Because the smaller the volume of the package, the more likely the concentration of the antibody is affected by evaporation and wall adsorption.

(3) If the reconstituted antibody is not used at one time, keep the remaining mother liquid at 4 degrees to avoid freezing again!

(4) The antibody working solution should be used up on the same day of the day, and should not exceed 1 day at 4 degrees .

(5) Never avoid storing antibodies in an automatic defrost refrigerator. Try to keep the antibody in the middle of the refrigerator instead of the door.

3. Most of the short-term storage of 4 weeks after antibody reception for 1-2 weeks has no effect on antibody activity.

If the antibody is used very quickly (1-2 weeks ) , it is recommended to store it at 4 degrees in order to avoid damage to the antibody activity by repeated freezing and thawing.

If you want to save for a long time, it is best to -20 degrees or -80 degrees. The most important point is to save the antibody correctly according to the method recommended in the manual!

However, products in the form of ascites should be frozen immediately after receipt! Because this kind of product contains a lot of proteases, long-term preservation at 4 degrees will lead to degradation of antibodies!

4. Transportation conditions of most antibodies: The general transportation process takes 1-2 weeks, so it is completed under 4 degrees.

The main purpose of 4 degree transportation is to avoid the damage of antibody activity caused by repeated freezing and thawing. ( If using dry ice transportation, the antibody is frozen when received, and needs to be thawed for dispensing. This is an additional freeze-thaw process. ) . Therefore, dry ice transportation is absolutely avoided during transportation!

Special antibody storage conditions:

1. Enzyme-linked antibodies: Always store at 4 degrees to avoid freezing. Otherwise it will lead to a decrease or loss of enzyme activity

2. Coupling antibodies: All conjugated antibodies are stored in brown tubes or protected from light using tin foil. In particular, fluorescent antibodies are extremely sensitive to light, so they are also protected from light in all experimental stages!

3. Isotype control of IgG3 : Always store at 4 degrees to avoid freezing. Because if the freezing and thawing process occurs, the antibody is very easy to form a polymer.

Whether it is preservation or transportation, absolutely avoid freezing and thawing!

Repeated freezing and thawing can cause antibody denaturation, resulting in the formation of multimers, thereby reducing the binding capacity of antibodies!
