J200 FS LA-LIBS System for Spatial Distribution of Plant Leaf Elements

Foreword
The spatial distribution analysis of plant leaf elements can be used in many applications such as plant physiology research, plant nutrient element transport analysis, plant enrichment research, etc. Especially the stratification of fresh leaves is of great significance. However, the conventional elemental analysis methods often require acid hydrolysis of the leaves, which cannot be analyzed as it is, and the bottleneck of the development of many scientific research fields. In this study, we used J200 343nm femtosecond LA-LIBS-ICP-MS to study the spatial distribution of trace elements in fresh leaves.

1 , multi-layer depth element distribution analysis



In this study, the leaf sample was delaminated in three layers. The results show the distribution of different elements at different levels. At the same time, LIBS can provide the distribution of Mg, Ca, C, H, O, N, F, Cl and major elements. These elements are not easily detected by ICP-MS, so we measured these elements using the LIBS module in the J200 343nm femtosecond LA-LIBS composite system. At the same time, the nanosecond LA-LIBS system may be able to achieve single layer erosion of plant leaves due to the large laser energy, but it is difficult to analyze and study multilayer erosion.

In this study, the MS signal intensity of Sr element in leaf sample 1 was analyzed. It was found that the distribution of Sr and B elements (measured by LA-ICP-MS) and Mg and Ca elements (measured by LIBS) were different under the surface layer and the surface layer. .


2 , healthy leaf and non-healthy leaf element content comparison

This study separately analyzed the spatial distribution of two different leaf elements in healthy leaves (sample 2) and yellow leaves (sample 3). It was found that the content of Mg in green healthy leaves was higher than that of Mg in unhealthy yellow leaves. The content of Sr; the elemental distribution of Sr is just the opposite. The K and Rb contents are higher in the veins, while the Sr elements are distributed in the leaves. In this study, the distribution of H elements in the two leaves was analyzed by the spectral acquisition module of J200 fs LA-LIBS system. It was found that the yellow leaves had high content of H elements in healthy leaves and could provide information of H elements at the same time.

to sum up
The J200 fs LA-LIBS system enables high-resolution elemental or three-dimensional spatial distribution analysis of fresh leaves of plants. This experiment requires proper parameter setting and airflow control experience, otherwise the sample is easily broken down by laser. Without good energy control and stability control, it is difficult to ablate and complete the experiment layer by layer, and the J200 fs LA-LIBS system perfectly completes the analysis. At the same time, the data analysis software provided by the J200fs LA-LIBS system can quickly analyze the spectral signals and MS signals, and quickly provide the required experimental results. The J200 fs LA-LIBS system will have great application potential in plant physiology research, atmospheric environmental pollution research, crop nutrition operation, and plant heavy metal enrichment research.
